Instruction for Mycelium germination from spores You can very easiely germinate spores on an agar medium. The so growen mycelium is called "Multi-Culutre" as opposite to a clone. With this methode of mycelium germination you will not get genetic identical copies of a mushroom culture - form the spores there will grow new organism of the same species. Mainly self made spore prints may contaminted with any bakteria, so we recommend to use antibiotic agar medium for spore germination. It is also possible to make mycelium like described in this instruction form spore syringes. Recommended Tools: (for product information, please click on product name) Spores (Sporeprint or Sporesyringe) Agar medium in petri dishes Oneway Inoculation loop or Needelholder with inoculation loop Parafilm to seal the petris Spiritus Lamp Facemask and Hairnet Latex gloves Workspace desinfection Handdesinfection Glove Bag (sterile workspace) For detailed instructions how to produce agar medium in petri disches by your self, please click here! General Procedere: To keep other microorganism (bakteria, mold,...) away from the agar medium, we recommend to use a HEPA-filter or a Glovebox / Inoculationbox. Spring clean and desinfect workspace and your hands. For reduction of contamination risk it is recommended to wear hearnet, facemask and latex gloves. Take the inoculation loop and rub it over the sproreprint to grab a few spores (rub till the loop gets a black spores veil). If you use reusable metal inouculation loop, you have to heat it over a spiritus lamp (put it into the flame till the forefront begins to glow). Let it cool down to room temperature bevor bringing in contact with the spores. Do not touch the spores, inoculation loop or agar medium with your fingers or bring in contact with any contamination resource! Now dispens the spores you have grabed with the inoculation loop on the agar medium. Draw with the loop a "S" from spores on to the agar medium. To reduce contamination risk please work fast and open the petri dish only as far as it is needful to get the inoculation loop in. Seal the petri dishes immediately with parafilm after you have dispensed the spores. If you use spore syringes, please dispens a few drops (1-2 ml) of spore solution on each petri dish. We reccomend to make about 5 petri dishes from each spore print, because not all spores will germinate equal. During the mycelium grows from the spores, store the petri dishes at the right temperature according to the growing conditions for the species you cultivate. After a few days up to 1 week the spore germination will get as strong that you can see it with the naked eye. For optimal growth we recommend to select the rhizomorph mycelium strains, before you inoculate grain or substrate with the mycelium.
